Put your eyeballs on it.. is the screw itself stripped, or the hole? You 100% sure the rotor is seated properly before trying to tighten it down? If you loosen the one screw that tightened, are you able to get the other one started? If you tighten one all the way it might have cocked the rotor lopsided preventing you from starting the other screw.
If the screw hole got stripped then you can either hack it by tapping it out for the next size screw, or replace the distributor ($$)... or you could simply try some threadlocker (blue) on the screw to see if it will hold.
If you're gonna tap the screw hole to the next size then you might consider removing the cam sensor first, easy to do, so that any filings won't get stuck to it. Fish 'em out with a magnet before re-installing the sensor.
