Very persistent misfire, help

I'm beginning to suspect something in the computer for my issue. Only #6 misfires and no other code

What does the scan tool show for MAP sensor info, O2 sensor info, and short- and long-term fuel trim data?

Have you tested for spark at the spark plug end of the plug wire, using a spark tester calibrated for HEI ignition?

Is the spark plug worn, fouled, carbon-tracked, cracked, or gapped improperly?

If you have compression, spark and a decent spark plug, and that cylinder still doesn't run, you've likely got a failed fuel system (such as a dead injector, including a failed or disconnected wire harness to the injector.)
