I installed my 1994 k2500 nv4500 transmission and transfercase in my 1997 k3500. The speedo doesn’t work. Should I get one for a 1997 or should it make no difference and maybe the 1994 speed sensor is bad?

I would think a difference in teeth ( if there were any) would be a difference in accuracy not whether it works or doesn’t.Does the VSS generate a signal at all? A multimeter with AC volts--or better-yet, a frequency counter--can verify that.
There may be damage to the wire harness carrying that AC voltage, or there may be differences in the reluctor on the transfer case output shaft. I don't know how many teeth the '94 has compared to the '97.
I don’t have a 97 transfercase, it’s from the 94, it should be the same as the 97 minus the sensorBoth the NV4500 and the 4L80 should have 6 bolt patterns, and 32 spline output shafts. Put your '97 t-case back in and the VSS should begin working again.

The old transmission and transfer case have been stole. I used the obd2 harnessBut you do have a 97 k3500. So what happened to the original tranny and t-case?
What wiring harness are you using? The OBD2 or the OBD1?
